Walk into any pharmacy or scroll any wellness feed and you’ll find shelves of anti-aging supplements promising to turn back the clock: collagen powders, NAD+ boosters, resveratrol capsules, astaxanthin softgels. Some of these have real human trial data behind them. Others are riding on animal studies, cell-culture results, or pure marketing. How to read supplement research (before you read the rest of this article)

Two things matter more than any single headline: who funded the study, and how many people were actually in it.

A 2025 systematic review and meta-analysis in the American Journal of Medicine looked at randomized controlled trials of oral collagen supplements for skin aging. Pooling all the trials together, collagen looked effective — better hydration, elasticity, and fewer wrinkles. But when the authors isolated only the high-quality studies and the studies not funded by supplement or pharmaceutical companies, the benefit disappeared (Myung & Park, American Journal of Medicine, 2025). That’s not a reason to dismiss collagen outright, but it’s a reason to be skeptical of any single flashy trial — and a pattern worth watching for across every supplement below.

Most human trials in this space also run 8-12 weeks with a few dozen to a few hundred participants. That’s enough to catch a real short-term effect on skin hydration or a blood marker. It’s nowhere near enough to prove a supplement slows aging or extends lifespan — that would require tracking thousands of people for years.

Collagen peptides: real short-term skin benefits, weaker at high quality

The short version: individual RCTs — including one with 119 women comparing 2.5g and 10g daily doses — have found real reductions in wrinkle depth and number after 8-12 weeks (Myung & Park, 2025). But as noted above, the effect shrinks or disappears in the subset of trials with no industry funding and the strictest methodology. If you want to try it, hydrolyzed collagen (5-10g/day) is low-risk and has more evidence behind it than most of what follows — just don’t expect dramatic results.

NMN and other NAD+ boosters: promising, but the trials are small and short

Nicotinamide mononucleotide (NMN) is one of the most hyped “longevity” supplements right now, based on its role in restoring NAD+, a coenzyme that declines with age and is involved in cellular energy production and DNA repair.

The actual human evidence is thinner than the marketing suggests, but it’s not nothing. A 2024 randomized, double-blind, placebo-controlled trial published in GeroScience gave 60 healthy older Japanese adults either 250mg/day of NMN or a placebo for 12 weeks. The NMN group maintained walking speed and had improved sleep quality compared to the placebo group, along with higher blood NAD+ levels (Morifuji et al., GeroScience, 2024). No serious side effects were reported.

That’s a genuinely useful data point — but it’s one trial of 60 people measuring walking speed and sleep, not skin aging or lifespan. There is currently no human trial showing NMN slows visible skin aging, and dosing, long-term safety, and effects beyond 12 weeks remain largely unstudied.

Resveratrol: strong in animal studies, unproven in humans

Resveratrol — the compound famous for showing up in red wine and grape skin — extended lifespan in yeast, worms, and some short-lived fish species in lab studies from the 2000s. That’s the origin of its “anti-aging” reputation. It hasn’t held up the same way in humans: resveratrol has poor bioavailability when taken orally, meaning very little of it actually reaches your bloodstream intact, and no human trial has been large or long enough to show it extends lifespan or measurably slows aging. Some trials do show modest improvements in metabolic markers like insulin sensitivity, but the headline “reverses aging” claim isn’t supported by current human data.

Astaxanthin: small trials show UV protection, but sample sizes are tiny

Astaxanthin is an antioxidant derived from algae, marketed heavily for skin and UV protection. The best human evidence is a randomized, double-blind, placebo-controlled trial in Nutrients (2018): 23 healthy participants took 4mg/day of astaxanthin or a placebo for 10 weeks, then had areas of skin exposed to UV light. The astaxanthin group showed a higher minimal erythema dose (meaning more UV exposure was needed to cause redness) and less UV-induced moisture loss than placebo, along with reduced wrinkle severity and improved elasticity over the study period.

The direction of the result is encouraging, but a 23-person, 10-week trial is a starting point, not proof. Larger, longer replication is still needed before “astaxanthin protects your skin from the sun” can be stated as settled science — and it’s not a substitute for actual sunscreen and the other daily habits that have far stronger evidence — including the ones covered in our piece on sleep and skin aging.

Vitamin C: the unglamorous one that actually matters

Vitamin C doesn’t get the buzzy “longevity” marketing that NMN or resveratrol do, but it has the most basic biochemical case of anything on this list: it’s a required cofactor for the enzymes that synthesize collagen in your skin. Diets low in vitamin C are associated with poorer collagen production and slower wound healing, and several of the stronger collagen-peptide trials paired the collagen with vitamin C rather than testing it alone. Getting enough (through citrus, peppers, or a basic supplement if your diet is low in it) is one of the least speculative things on this list — it’s supporting an established biological pathway, not chasing a new mechanism.

What to skip or treat with real skepticism

  • Biotin for skin/hair “anti-aging” — genuinely helpful if you’re one of the rare people with a biotin deficiency; there’s no good evidence it does anything for skin aging or hair in people who aren’t deficient.
  • “Detox” or “cellular renewal” blends — proprietary multi-ingredient blends rarely have the blend itself tested in a trial; each ingredient may have some individual evidence, but the specific combination and dose usually doesn’t.
  • Mega-dose antioxidant stacks — more isn’t automatically better; some trials of high-dose antioxidant supplementation (unrelated to the specific ones above) have shown neutral or even mildly negative effects on health markers.

Bottom line

If you want to spend money on a supplement with the most human evidence behind it for skin aging specifically, hydrolyzed collagen (with vitamin C) and astaxanthin have the most direct skin-focused RCT data, with the caveat that collagen’s effect shrinks in the highest-quality, non-industry-funded trials. NMN has real short-term data on walking speed and sleep in older adults, not skin. Resveratrol’s human anti-aging case is currently the weakest of the group despite being one of the most marketed.
